-- TH1: CoUNc]L FoR TOBACCO RESLAUC•11-U.g.A., INc. February 15, 1979 1 ' MEMORANDUM I I I I f T0: Drs. Hockett, Jacobson, Stone and Wyatt FROM: W. U. Gardner SUBJECT: Application 1266. John E. Repine, M.D., Webb-Waring Lung Institute, Denver, Colorado. "The Role of Phagocytes in the Development of Centrilokular Lmphysema." The applicants are both M.D.s from the University of Minnesota, 1971 and 1969, Repine and Hoidal respectively. Beginning in 1979 Repine is Assistant Director of Webb-Waring and Associate Professor of Medicine, University of Colorado. Hoidal seems to be at Minnesota as there is no position designated after 1975. The papers and preprints submitted all carry Minnesota origins. They have pending applications to the Heart Association and NIH for $70,000 for work on 02 toxicity in lungs. They are asking for a part-time technician and animal caretaker. Other fmding is for consumables and animal care and purchase. All in all the request seems reasonable. Projection. The major goal is to determine the role of lung phagocytes in the development of centrilobular emphysema. PMA from smokers (CS) and nonsmokers (NS) will be followed for their capacity to attract monocytes and PMNs, whether there are differences in 02 uptake, suoeroxide (02-) anion H202, hydroxyl radical superoxide dismutase and catalase difference in CS or NS. Cd C12 given chronically will be used in rats to produce a CLE condition. CdC12 will be administered as a vapor. It is stated that Cd decreases superoxide dismutase (SOD) and catalase which are important in 02 and H20L inactivation. Animals are said not to develop CLE when exposed to smoke. The first studies will be to determine the levels of a possible chemotactic factor in the PAMa of smokers and NS that attract PMNs and monocytes. 02, H202, OH, SOD and catalase, malondiaNeWehyde activites and Cd content will be determined in young persons. Rats exposed to CdC12 and other toxic substances will be studied for the activities mentioned above. Dr. Hoidal reportedly has done over 2,000 lavages. Young CS and NS will be used because they want to study the early st age response to smoke. The applicants have had experience using most of the techniques required for both the morphological and biochemical studies. This application impresses me in that it relates to free radical influences and combines both human and laboratory animal studies. Furthermore the combined talents of two young investigators will be applied to the :;tudies. This should complQment other studies on COLD. W. U. Gardner WUG:ek
