BC3FI/Cum female mice werc exposed daily by "nose-only" inhalation to whole smoke from 2 reference cigarettes: 2R1 and 3A1, of varying tar/nicotine content. Under standard smoking conditions using the SEM IIB smoking machine, '6300 Ug total particulate matter was deposited/lung/day. We per- formed ].abelling Ind^x estimations :'n lung as an indicator of localized smoke-induceli effects, and studied SCE's in hone marrow as an indicator of systemic smoke-induced eifects. When mice were pulsed briefly with 31l-tllymidine immediately after the last smoke exposure, and the lung analyzed autoradiographically, DNA synthesis was found to have increased compared to controls after 9 weeks' exposure and Qrc increase persistcd for 52 weeks. Maximum response was found at 13 weeks, when thc percent labelled cclls were 'L2CX (2R1) and til4X (3A1) greater than controls. Labelling was confined to alveolar cells; none were found in hror.chi'. The majority (ti67X) of labelled cells contained less than 30 grains/nucleus suggesting that in addition to replicative D14A synthesis, repair synthesis occurred as well. Analysis of the bone marrow of mice revealed that 1, 4 or 46 week- exposure to smoke from either cigarette type resulted in N8 SCE's/cell compared to 'L4 for the unexposed or control sham- exposed mice. When the mice wF.re withdrawn from smoke ex- posure after L week or 46 weeks a significant increase in SCE's persisted for at least 1 week.
