Using the SEM IIB cigarette smoking system, "nose-only"

exposure of BC3F1/Cum mice to 2R1 cigarettP smoke (high tar,

high nicotine) and 3A1 cigarette smoke (high tar, low nicotine)

OS resulted in deposition of 100-400 ug smoke particulates/mouse/
1k. day, with '6905 of the deposition in the respiratory tract.

Deposition of smoke particulates in the lung of these mice re-
sulted in: a) 2-4 fold increase in lung microsomal mono-
oxygenases by 6 hrs post exposure, b) '~2 fold increase in lung
1: ornithine decarboxylase activity after 12 weelcs smoke exposure
1!
(5 days/week), c) 10-20 fold increase in 3H-thymidine incorpo-
10
ration (i.e. DNA, synthesis rates) by 9 weeks of daily smoke
exposure, d) ti309~ increase in lung/body weight ratios (and
total lung protein) by I week smoke exposure, and e) '~2 fold
increase in sister chromatid exchanges in bone marrow after 1
week smoke exposure. Exposure to 3A1, but not 2R1, cigarette
smoke resulted in an ti507 decrease in pulmonary DNA repair
capacity. Exposure to smoke from either the A or R series
cigarettes for ti52 weeks failed to alter splenic B-cell re-
x
sponses (Jernc plaque assay). Thus, cigarette smoke induces
both organ specific and systemic biological responses in a
murine model system. These responses could form the basis
o
~~ for comparing the potential effects of various cigarette types
02 and smoking conditions.
